♫ Sunday, July 3rd, 2011International Stem Cell Corporation (OTCBB:ISCO) (http://www.intlstemcell.com), the first company to perfect a method of creating human "parthenogenetic" stem cells derived from unfertilized human eggs, reports that its wholly owned subsidiary, Lifeline Cell Technology (Lifeline), grew 2011 first quarter product sales by 35% and gained well over 200 new customers compared with the prior year through new product introductions and the development of worldwide distribution channels. Lifeline also made significant progress in its strategy to expand its product applications into manufacturing human tissues and cells for clinical use. Lifeline Cell Technology develops, manufactures and markets the Lifeline® brand of cell culture products used by researchers to grow human cells for basic and pre-clinical research. Lifeline also fulfills an important role in parent ISCO's long-term strategy to be a leading developer and manufacturer of human cells and human-cell-based products for clinical applications.
Lifeline's chief executive officer, Jeffrey Janus, stated, "Our product sales growth was largely due to the development and launch of more than 30 new products in 2010, including products that allow researchers to study human stem cells. Lifeline also opened new distribution channels in Japan, India, Taiwan, South Korea and Singapore and has achieved significant sales in those new markets. Importantly, we have also moved closer to validating the feasibility of ISCO's strategic plan as our proprietary FibroLife® media is being used to cultivate tissue-engineered blood vessels for Cytograft Tissue Engineering in Novato, California. Cytograft's vessels are expected to be used as coronary grafts for coronary bypass procedures, as peripheral grafts to prevent lower limb amputations and as living tissue shunts for hemodialysis patients."

Kurt May to Join International Stem Cell Corporation as Senior Vice President

Andrey Semechkin, Ph.D., CEO of International Stem Cell Corporation (OTCBB:ISCOE) ("ISCO") announced today that Kurt May, a highly experienced senior business executive and biotechnology entrepreneur, is concluding his service as a faculty member at the University of San Diego's School of Business Administration and is joining ISCO as a Senior Vice President.
Mr. May's duties will include mergers and acquisitions and development of new international collaborations, and he will work directly with Dr. Semechkin in implementing the Company's business model of combining cutting-edge scientific research and building multiple revenue streams to support that research.
Prior to joining ISCO, Mr. May was a senior executive with GTE Corporation, a Fortune 50 company, where he enjoyed 23 years of progressive management experience. Mr. May then went on to serve as Executive Vice President and Chief Operating Officer with PriceSmart Inc. Since 2001 Mr. May has founded and led a privately owned biotech company, Psynomics Inc., as CEO. Psynomics Inc. is a spin out of the University of California San Diego, where Mr. May served as a faculty member and Assistant Dean of the Rady School of Management from 2005 until 2009.

International Stem Cell Corporation Discovers Method to Produce Uncontaminated Liver Cells: New Platform to Produce Various Cell Types Free From Undifferentiated, Potentially Tumorigenic, Cells
♫ Sunday, June 19th, 2011International Stem Cell Corporation (OTCBB:ISCOE) announces the discovery of a novel, patent-pending technology to produce unlimited numbers of liver cells (hepatocytes) that are free of contamination with potentially dangerous undifferentiated cells. The technology is based on the natural, physiological properties of the cellular environment, and does not require any additional purification of the final product. The starting materials for the production are pluripotent stem cells, either ISCO's proprietary human parthenogenetic stem cells (hpSCs) or human embryonic stem cells (hESCs).
The technology is described in a new article that will appear in Cell Transplantation, the Regenerative Medicine Journal (currently the article is available only in electronic form). The published data also reinforce that hpSCs can provide the cellular material necessary for the implementation of cell-based therapies.
Marie Csete, M.D., Ph.D., co-author of the paper, said, "Derivation of differentiated cell products that are not contaminated with undifferentiated cells solves a major technical roadblock in the development of all pluripotent stem cell-based therapies because undifferentiated stem cells with tumorigenic potential can persist through long differentiation protocols. Therefore methods to generate pure differentiated cells for transplantation are critical for creating successful cell therapies. Furthermore, the elegant technology developed by ISCO scientists to enforce critical steps in differentiation illustrates the power and importance of basic engineering tools in stem cell biology."
Nikolay Turovets, Ph.D., Director of Research and Therapeutic Development at ISCO and co-author of the paper stated, "The technology discovered by our research team is based on reproducing features of the normal human embryonic microenvironment. The method uses a differentiation device that incorporates a three-dimensional extracellular matrix, combined with a porous membrane. Treatment of undifferentiated cells above the membrane using differentiation-directed proteins results in permitting the desired cells to migrate through the membrane into the matrix, where they further differentiate into functional hepatocytes."
Jeffrey Fair, M.D., liver transplant surgeon and Director of Translational Research for the Cedars-Sinai Comprehensive Transplant Center and Department of Surgery in Los Angeles, and co-author of the paper said, "Derivatives of HLA-homozygous hpSCs are likely to be significantly less susceptible to immune rejection after transplantation, thus offering major advantages over other pluripotent cell sources of hepatocytes. In the paper, we demonstrated that hepatocytes derived from hpSCs in this differentiation system demonstrate activities that are missing in a number of metabolic liver diseases, such as expression of AAT and OTC genes. Thus, personalized HLA-matching, as well as presence of the required activities make the pure hepatocyte population derived from hpSCs an attractive development candidate for cell therapy of metabolic liver diseases, urea cycle disorders, AAT-deficiency or other liver diseases in which a single hepatocyte product can ameliorate disease."
The approach used to derive a pure hepatocyte population is a technology platform that may allow derivation of various cell populations from different sources.
Andrey Semechkin, Ph.D., President and CEO of ISCO, and co-author of the paper stated, "We have discovered technology that has a number of features and advantages, and our plans are to expand the applications of this platform to develop new products. This system is universal and does not depend of the particularities of cell lines. As such it could be successfully used with hpSCs, hESCs and, we believe, with induced pluripotent stem cells (iPSs). Because isolation of undifferentiated cells happens at the first step of the differentiation procedure, the technology could be used to isolate other cell types, for example pancreatic or heart cells. Also, because technology is based on a natural process, cell separation is accomplished without any cell damage, in contrast to physical purification methods such as FACS, or magnetic sorting."

International Stem Cell Corporation Reports First Quarter Revenue of $1.5 Million on Strong Initial Sales of Skin Care Products and Strong Growth of Research Product Sales
International Stem Cell Corporation (OTCBB: ISCO) (ISCO) announces that revenue in the first quarter of 2011 was $1.5 million, compared with revenue of $273,000 in the first quarter of 2010. The increase was primarily due to $1.1 million in sales of Lifeline Skin Care products achieved in the pilot direct-to-consumer launch. Adding to the revenue growth was a 35% increase over the prior year in sales of human cell culture research products from subsidiary Lifeline Cell Technology.
ISCO also disclosed that it is delaying the filing of its Report on Form 10-Q for the period ending March 31, 2011 in order to resolve an accounting question as described below.
Shortly before the scheduled date for filing the 10-Q, ISCO identified an issue related to whether a technical interpretation of certain accounting guidance found in FASB ASC Topic 815, Derivatives and Hedging could require a change in the manner ISCO has historically accounted for the value of warrants originally issued as compensation to brokers working for the placement agent in the Company's 2007 initial public offering.
The accounting question is highly technical in nature and would have no impact on cash or revenues, but relates solely to the non-cash accounting for warrants issued several years ago. Rather than publish information that might be subject to further revision, ISCO has elected to delay the filing of its 10-Q until a decision can be made as to whether or not adjustments will need to be made and, if so, exactly what the accounting entries will be. The Company is committed to resolving this question as promptly as possible and will disclose the conclusion in the 10-Q.

Scientists at the University of California, San Diego, announced last week that they had discovered that the class of stem cells known as "induced pluripotent stem cells" or "iPS" cells could cause an immune rejection problem when transplanted into mice. Scientists had hoped that these cells would have two big advantages over embryonic stem cells: they would not be controversial because their creation did not entail the destruction of human embryos; and, since the stem cells could be made from a particular patient’s skin cells, they could be used to make tissues that would not be rejected by the patient’s own immune system. Although responses in humans may be different, the discovery of an immune response in mice suggests that the second of these hopes may not be so easily realized.
That is disappointing news in many ways, but it illustrates what we at International Stem Cell Corporation (ISCO) think will be one of the great benefits of the Parthenogenetic Stem Cells our scientists have created and patented.
Parthenogenetic stem cells are not only pluripotent they also do not involve any destruction or damage to a viable human embryo. Since they are never fertilized (and can't become a child) they carry the DNA of only the egg donor and not the added DNA of a father or other sperm donor. This results in the possibility of matching these cells to large groups of people without causing immune response problems.
The science is a bit complicated, but the result is illustrated by the fact that the very first stem cell line ISCO created using its newest techniques has the potential to match the immune systems of over 50 million people. With the right donors, cell lines from as few as 50 donors (one respected scientist has said as few as 10 donors) could match a very large portion of the world's population. There is a lot of work yet to be done, but if one thinks of it as comparable to a blood bank, but for stem cells instead, the concept becomes quite exciting.
Lastly, even if iPS cells are never used for transplant therapy, they still have wonderful research potential, so those who hoped for medical breakthroughs using them should not despair. They are, and will be, very important, even though there are better pathways to transplant therapy.

International Stem Cell Corporation (OTCBB: ISCO) announced today that two of its leading scientists, Vice President Dr. Ruslan Semechkin and Director of Research and Therapeutic Development Dr. Nikolay Turovets, will present the results of their most recent experiments on the therapeutic use of human parthenogenetic stem cells (hpSCs) at the 14th Annual Meeting of American Society of Gene and Cell Therapy in Seattle.
ISCO's breakthrough discoveries have resulted in a new type of pluripotent human stem cells with distinct advantages over other human pluripotent stem cells. ISCO uses unfertilized oocytes to create human "parthenogenetic" stem cells. Like embryonic stem cells, hpSCs are pluripotent, i.e. they have the capacity to become almost any cell type in the body, yet avoid ethical issues associated with use or destruction of viable human embryos. The data presented at the Seattle conference represents progress made in two specific areas: 1) the successful derivation of neuronal stem cell lines, which can potentially be used for treatment of degenerative diseases of the central nervous system (CNS), and 2) the differentiation of hpSCs towards liver cells and their progenitors.

Following the highly successful launch in late 2010 of Lifeline's first two products with sales that greatly exceeded expectations, Lifeline has now commenced a new phase in the marketing plan for its skin care products.

Good morning to all our shareholders and friends of ISCO. Let me make just a few comments about where we are and the progress we have made over the last year. All of the developments and news I will discuss were announced in the past 12 months, but with the passage of time it is easy to lose track of how much progress ISCO has actually made. I won’t mention everything—we don’t have that much time, but some highlights will, I hope, help put these recent achievements into perspective.
A little less than a year ago, in June 2010, our first parthenogenetic patent application was formally approved by the US Patent Office. More applications are pending, but this approval established ISCO as the lawful owner of the rights to produce human stem cell lines through parthenogenesis. That is a platform on which we expect to build for a long time.
Also in June we eliminated all of our outstanding corporate debt and we remain debt free.
In July we announced the signing of a distribution contract with Sristi Biosciences, a major seller of research products in India, which continues the international commercial expansion of our Lifeline Cell Technology® brand.
In October we announced the first steps toward the formation of a major funded collaboration in India with Insight Bioventures India Private Limited (IBVI) to develop treatments for corneal damage and retinal disease. Planning for that work is ongoing and the Executive Director of Insight Bioventures has flown in from India to meet with us today, so we remain very optimistic about this project.
In November, we presented the results of scientific studies demonstrating a new and better method for differentiating our parthenogenetic cells into liver cells, a critical step toward using them to treat liver disease.
In November and December we launched the first test marketing runs of our new skin crème products, first to our own shareholders and friends, then to a select mailing list developed by our marketing partner, John Mauldin. Those resulted in sales of over 7,000 bottles of our new products.
In December we established $25 million financing commitment that provides access to capital on an as-needed basis over a three year period, but never requires the sale of stock unless we think it will benefit the company and its shareholders. This agreement provides us a high degree of flexibility in meeting our financial needs.
In January 2011 we announced publication of peer-reviewed studies further validating the functional equivalency of our parthenogenetic stem cells with embryonic stem cells. These studies confirm our ability to benefit from much of the millions of dollars of research on embryonic stem cells over the last decade. In short, we have a running start in the search for cures.
Throughout the year we have made steady progress in development of liver cells and liver precursor cells, culminating in an announcement in April of this year that we had successfully completed the first in a series of pre-clinical tests of parthenogenetically derived liver cells.
Last, but by no means least, we have received the necessary approvals for creating new Parthenogenetic Stem Cell Lines in the United States. These will be clinical grade lines suitable for human trials, and are the first major step in this country to begin building a “Bank” of stem cells enabling the matching of immune systems of millions of people worldwide, with the potential to eliminate or reduce the harmful effects of immune suppressing drugs that would normally have to be used with stem cell transplant procedures.

International Stem Cell Corporation Completes First Series of Pre-Clinical Testing of Hepatocytes Derived from Human Parthenogenetic Stem Cells

International Stem Cell Corporation (OTCBB:ISCO) (ISCO) announces successful completion of the first series of preclinical testing of hepatocytes derived in the lab from human parthenogenetic stem cells (hpSC). In the transplantation mouse model, inoculated cells were capable of engrafting and surviving in specific niches within the liver, and were further developing into cells with essential hepatocyte-like features. Moreover, the transplanted cells could be identified in recipient tissue for a prolonged period of time.
The findings of these studies will be presented at the annual meeting of American Society of Gene & Cell Therapy, May 18-21 in Seattle, and at the International Society for Stem Cell Research annual conference, June 15-18 in Toronto.
"These results mark the achievement of a key milestone in our preclinical research," said Andrey Semechkin, Ph.D., ISCO's Chief Executive Officer. The Company also announces the signing of a joint collaboration agreement with Cedars-Sinai Medical Center in Los Angeles to conduct research to develop therapies for liver diseases, in particular urea cycle disorders. Jeffrey Fair, MD., a liver transplant surgeon and Director of Translational Research for the Cedars-Sinai Comprehensive Transplant Center and Department of Surgery, will lead the Cedars-Sinai research team.
Dr. Fair said, "Urea cycle disorders are genetic deficiencies of liver function, which mostly affect newborns and oftentimes cause catastrophic neurological injury. It has been shown that transplantation of donor hepatocytes can save patient lives. Therefore, derivation of hepatocytes from hpSCs that can be immune-matched to the patient is a very pressing goal."
The Company also announces the completion of research focused on the investigation of cytochrome P450 activity and corresponding genes in hepatocytes derived from hpSC. According to the results, the differentiation technology developed by ISCO allows the creation of hepatocytes in the fetal stage of development. This research may contribute to the design of a product for future drug testing and discovery.

International Stem Cell Corporation Receives Important Approvals Towards Creating New Parthenogenetic Stem Cell Lines in the United States
♫ Sunday, March 13th, 2011International Stem Cell Corporation (OTCBB:ISCO), http://www.internationalstemcell.com, announced today the achievement of a major milestone in its efforts to establish a bank of immune-matched parthenogenetic stem cells in the United States by receiving the statutory approvals required to move forward with the next phase of its stem cell banking program.
An independent Stem Cell Research Oversight (SCRO) committee approved ISCO's program to obtain human oocytes (eggs) in California. The independent SCRO committee is responsible for reviewing the scientific rationale, ethics and compliance with State and Federal regulations and guidelines for studies involving human stem cells. This approval in combination with the earlier reported Institutional Review Board (IRB) clearance with regard to medical safety opens the way for ISCO's stem cell banking program to begin registering clinical investigators. ISCO's study is designed to produce new clinical-grade human parthenogenetic stem cell lines that can be immune matched to millions of individuals for use in the emerging field of regenerative medicine.
ISCO's breakthrough discoveries have resulted in unique stem cells carrying distinct advantages over other human pluripotent stem cells. ISCO uses unfertilized oocytes to create human "parthenogenetic" stem cells (hpSCs). Like human embryonic stem cells (hESCs), hpSCs are pluripotent, i.e. they have the capacity to become almost any cell type in the body, yet avoid ethical issues associated with use or destruction of viable human embryos. Unlike hESCs, hpSCs can be created in a form such that they can be immunologically matched to millions of individuals.
Currently ISCO has a collection of ten hpSC lines that are shared with collaborators and used for research purposes. Although these ten stem cell lines conform to all relevant US standards, they were derived outside the United States. New cell lines created under this study will be ISCO's first hpSC to be produced in the United States. ISCO's scientists, leaders the field of creating therapeutic cells from hpSC, will work with these lines targeting new treatments for human diseases. The company will also share these cells with other scientists to further clinical research. ISCO intends to derive and store these cells at its Oceanside facilities, which are designed to create cells suitable for clinical use.
